SUM159 and WHIM12 cells were reverse transfected with Dharmacon SMART siRNA pools in 6 well dishes. For SUM159 cells, 60 hours post transfection, cells were treated with either vehicle or 10ng/mL TGFB for 3 hours. Total RNA was collected from SUM159 cells sixty three hours post transfection. Total RNA was collected from WHIM12 cells forty eight hours post transfection.

SUM159 and WHIM12 cells were grown in the F12, (Gibco) and MEM (Gibco), respectively, and split 1:3 every two days.

The WT Expression HT Kit for Robotics (Ambion) was used to generate sense-strand cDNA from total RNA. Following synthesis of sense-strand cDNA, the cDNA was fragmented and labeled with the Affymetrix GeneChip HT Terminal Labeling Kit.

Fragmented and labeled cDNA was used to prepare a hybridization cocktail with the Affymetrix GeneTitan Hybridization Wash and Stain Kit for WT Arrays.

| Scan protocol |
Hybridization, washing, staining and scanning of the Affymtrix peg plate arrays was carried out using the Affymetrin GeneTitan MC Instrument.
