SUM159 and WHIM12 cells were reverse transfected with Dharmacon SMART siRNA pools in 6 well dishes. For SUM159 cells, 60 hours post transfection, cells were treated with either vehicle or 10ng/mL TGFB for 3 hours. For WHIM12 cells, 48 hours post transfection, cells were treated with either vehicle or 10ng/mL TGFB for 3 hours.

SUM159 and WHIM12 cells were grown in the F12, (Gibco) and MEM (Gibco), respectively, and split 1:3 every two days.

Fragmented and labeled cDNA was used to prepare a hybridization cocktail with the Affymetrix GeneTitan Hybridization Wash and Stain Kit for WT Arrays.

| Scan protocol |
Hybridization, washing, staining and scanning of the Affymtrix peg plate arrays was carried out using the Affymetrin GeneTitan MC Instrument.
