RNA mini kit (Qiagen) mRNA was isolated from total RNA (500ng) using the Ambion's mRNA direct kit (Dynabeads® mRNA Purification Kit ). Libraries were prepared with the Ion Total RNA-Seq Kit v2 according to the manufacturer's instructions. ERCC were spiked-in before the mRNA isolation.
