Treatment protocol We sampled skeletal muscle biopsies from vastus lateralis with a Bergstrom needle using suction and local anaesthesia (1% lidocaine) at both visits (B and C). Biopsies were snap-frozen in liquid nitrogen and stored at -80°C subsequently.
Growth protocol 16 severely obese patients without type 2 diabetes mellitus (OB) and nine severely obese patients with type 2 diabetes mellitus (T2DM) were elected for Roux-en-Y gastric bypass surgery. We examined the participants after a diet-induced weight loss (B) and approximately 4 months post-surgery (C).
Extracted molecule total RNA
Extraction protocol Total RNA was extracted from approximately 20 mg of muscle tissue using Trizol (Invitrogen).
Label SYBR green
Label protocol 30 ng RNA was reverse transcribed in 30 μl reactions using the miRCURY LNA Universal RT microRNA PCR, Polyadenylation and cDNA synthesis kit (Exiqon). cDNA was diluted 100x and assayed in 10 ul PCR reactions according to the protocol; each microRNA was assayed once by qPCR on the microRNA Ready-to-Use PCR, Human Panel I using ExiLENT SYBR® Green master mix. Negative controls excluding template from the reverse transcription reaction was performed and profiled like the samples. The amplification was performed in a LightCycler® 480 Real-Time PCR System (Roche) in 384-well plates.
