Growth protocol |
Cell lines were maintained in Dulbecco's Modified Eagle Medium (DMEM) (Invitrogen, Carlsbad, CA) supplemented with 10% fetal bovine serum (Gemini, Woodland, California), 2 mM L-glutamine, 1 mM sodium pyruvate, 0.1 mM non-essential amino acids, 5 mg/ml penicillin-streptomycin (Invitrogen, Carlsbad, CA) and kept at 37ºC incubator with 5% CO2. Cells were transferred to 10% charcoal-dextran-treated, heat-inactivated fetal bovine serum for 48 hrs prior to hormone treatment.

RNA was extracted from cells either vehicle treated or treated with 100 nM hormone (dexamethasone, Dex) for 0h, 2h, 4h or 8h. 3134 and AtT-20 RNA for microarray analysis was prepared via standard manufacturer protocols (Qiagen, Valencia, Ca) using cells resuspended in Trizol reagent (Invitrogen, Carlsbad, Ca)
