Extraction protocol RNA was extracted with the RNAqueousTM-Micro Total RNA Isolation Kit AM1931 (Thermo Fisher Scientific K.K., Yokohama, Japan) according to the manufacturer's instructions.
Label biotin
Label protocol Aliquots of total RNA (50 ng) isolated from biopsied liver tissues were subjected to amplification using the Ovation Pico WTA System V2 3302-12 (AS ONE, Japan) according to the manufacturer’s instructions. Approximately 10 μg of cDNA was amplified from 50 ng of total RNA, and 5 μg of cDNA was used for fragmentation and biotin labeling using the FL-Ovation cDNA Biotin Module V2 (NuGen) according to the manufacturer’s instructions.
 
Hybridization protocol Biotin-labeled cDNA was suspended in 220 ul of hybridization cocktail (NuGen) and 200 ul was used for hybridization to the Affymetrix Human 133 Plus 2.0 microarray chip containing 54,675 probes.GeneChips were washed and stained in the Affymetrix Fluidics Station 400.
Scan protocol After stringent washing, the microarray chips were stained with streptavidin-phycoerythrin and probe hybridization was determined using a GeneChip Scanner 3000 (Affymetrix).
Data processing The data were analyzed with Microarray Suite version 5.0 (MAS 5.0) using Affymetrix default analysis settings and global scaling as normalization method. If the detection call contained 'Absent', the probe set was removed from analysis.
