Robust hepatitis E virus infection and transcriptional response in human hepatocytes

(Submitter supplied) In this study, we established a protocol based on the HEV genotype 3 p6 (Kernow C-1) and the human hepatoma cell lines HepG2 and HepG2/C3A with different media conditions to produce HEV cell culture-derived particles (HEVcc) with viral titers between 10e5 to 10e6 FFU/mL. Viral titers could be further enhanced by an HEV variant harboring a mutation in the RNA-dependent RNA polymerase. These HEVcc particles were infectious in primary human hepatocytes. more...

Real-time quantitative PCR analysis of human HepaRG cells infected with hepatitis E virus (HEV) [pre-array]

(Submitter supplied) We aimed to identify interferon (IFN)-regulated genes that are differentially expressed during chronic HEV infection in human hepatocytes, the main site of HEV replication, using HepaRG cells. We have performed a preliminary screen using whole-cell RNA extracts prepared from HepaRG mock-infected or infected cells to determine whether HEV was able to trigger an IFN response. Different multiplicities of infection (MOI) (10 and 100 genome equivalent (GE)/cell) and time points (D+7, D+14, D+26, D+40, D+72 and D+100) were analyzed using the PCR array. more...

Targeting cholesterol metabolism as efficient antiviral strategy against the Hepatitis E virus

(Submitter supplied) We used whole genome expression profiling to analyze the effects of Hepatitis E Virus (HEV) on the mRNA transcriptome of A549/D3 cells - particularly to identify differentially expressed genes which are associated with cholesterol metabolism.

Transcriptional profiling of stem cell-derived polarized hepatocytes

(Submitter supplied) Human stem cell-derived hepatocyte-like cells (HLCs) offer an attractive platform to study liver disease and to test drugs. However, despite their advantages over other hepatocyte culture systems, HLCs lack several in vivo characteristics including cell polarity, which is critical for proper hepatocyte biology. We report a stem cell-based differentiation protocol that uses transwell filters to generate columnar polarized HLCs with clearly defined basolateral and apical membranes separated by tight junctions. more...
