Gene expression signatures of MDCK-pTR GFP-RasV12 cells cultured alone or mixed with MDCK cells

(Submitter supplied) Newly emerging transformed cells carrying genetic defects, including the active Ras mutant, are apicall extruded when surrounded by normal epithelial cells. This biological phenomenon is termed as cell competition. It remains elusive which factors prime RasV12-transformed cells to be expelled via cell competition. Here, We performe microarray analysis to search for molecules whose expression is changed in RasV12 cells surrounded by normal cells.

Gene expression signatures of MDCK-pTR GFP-RasV12 mCherry-β-catenin Δ131 cells cultured alone or mixed with MDCK mCherry-β-catenin Δ131 cells

(Submitter supplied) Normal epithelial cells exert their competitive advantage over RasV12-transformed cells and eliminate them into apical lumen via cell competition. However, it remains elusive which internal or external factors compromise cell competition and provoke oncogenesis. Here, we examine the effect of sequential accumulation of gene mutations mimicking multi-sequential carcinogenesis on RasV12-induced cell competition. more...

Development of gene signatures for doxorubicin resistance and cancer stem cell CD44+, CD24-/low ALD+ isolated from canine mammary adenocarcinoma cell line (mRNA)

(Submitter supplied) CMT-Stylo is a canine mammary gland adenocarcinoma cell line. From this cell line, a doxorubicin resistant subline called CMT-Star was developed by culturing the cells in the presence of doxorubicin from low concentration to 100 nM doxorubicin (to induce drug resistance). The CMT-Stylo cells were maintained in Dulbecco’s modified eagles medium (DMEM) supplemented with 10% Foetal bovine serum (FBS) and 1% Penicillin-Streptomycin (ThermoFisher Scientific, USA) in 5% CO2 at 370C. more...

Gene expression profile of small intestinal mucosa in dog repeatedly infected with the cestode Echinococcus multilocularis

(Submitter supplied) Previously, we found that the exclusion of adult worm occurred early stage of reinfection in dog repeatedly infected with E. multilocularis. In this study, we compared gene expression profile in small intestinal mucosa between control (normal dogs), first-infection group (dogs infected with E. multilocularis for the first time) and repeated-infection group (dogs repeatedly infected with this parasite 4 times). more...

Change in gene expression of a canine melanoma cell line caused by exposure to nonsteroidal anti-inflammatory drugs (NSAIDs) .

(Submitter supplied) Nonsteroidal anti-inflammatory drugs (NSAIDs) has been suggested as adjunctive anti-tumor agents in human and veterinary medicine. However, its anti-tumor molecular machinery is still controversial. Therefore, we performed whole transcriptome analysis to discover gene expression influenced by NSAIDs treatment.

Retinal Gene and Protein Expression Associated with CNTF-Induced Deconstruction of Photoreceptor Outer Segments in normal and CNGB3-achromatopsia dogs

(Submitter supplied) Background: Intravitreal injection of CNTF leads to deconstruction and regeneration of photoreceptor outer segments in normal dogs, and improves the success rate of viral CNGB3 gene replacement therapy in dogs (> 1 year) with CNGB3-achromatopsia. Objectives: The goal of this study was to examine the changes caused by intravitreal CNTF injection to the retinal gene expression profiles and the retinal function of normal and CNGB3-achromatopsia affected dogs. more...

The transcriptomic profile of peripheral blood nuclear cells in dogs with heart failure

(Submitter supplied) The aim of the study was to identify genes which are differentially expressed in the blood of dogs suffering from heart failure (HF) in comparison to healthy control dogs. The dogs with HF were categorized according to the ISACHC classification system (International Small Animal Cardiac Health Council).

H3N2 canine influenza virus causes severe clinical signs in dogs with induction of genes related to inflammation and apoptosis

(Submitter supplied) We demonstrated canine influenza virus (H3N2) pathogenicity to dogs using microarray analysis. Many genes related to innate immunity, such as toll-like receptors, immune cells of natural killer cells, macrophages, neutrophils, nitric oxide and reactive oxygen species, and interferon, were induced.

Acid Stress Increases Gene Expression of Proinflammatory Cytokines in Madin Darby Canine Kidney Cells

(Submitter supplied) Metabolic acidosis exacerbates chronic kidney disease, in part, by stimulating renal endothelin production. To determine if acidosis alters expression of other genes, we examined the effect of exposure of MDCK cells to pH 7.4 and pH 7.0 for 24 hours on gene expression using a canine derived microarray. Exposure to this pH stress for 24 hours led to increased expression of 278 genes (2.2% of the transcriptome) by at least two fold and 60 of these (21%) were upregulated by more than 3 fold. more...
